KinExA® Method

1) The KinExA 3000 automatically packs a fresh column of micro-beads
(coated with the ligand (L) of interest) for each sample run.
2) The sample containing ligand, free receptor (R), and ligand-receptor complex (LR) is pulled rapidly through the column to keep the contact time of the sample with the beads extremely brief. Free receptor is bound to ligand on the beads.
3) Secondary label such as Cy5 fluorescently labeled secondary antibody to the receptor is then passed through the column. Labeled secondary antibody binds to the bound receptor (R).
4) A buffer wash removes excess label, leaving fluorescence signal on the bead column directly proportional to the amount of free receptor in the original sample.
