ANTIBODIES

“To gain a more quantitative measurement of the antibody binding affinities to guide our epitope mapping experiments, we turned to KinExA analysis, a method that measures equilibrium binding affinities of unmodified molecules in the solution phase.”

— Wong X. et al., 2017. Fine epitope mapping of two antibodies neutralizing the Bordetella adenylate cyclase toxin. Biochemistry, 56(9), pp. 1324–1336

CELLS

“Having a robust methodology for characterizing antibody interaction with a membrane protein target is essential for providing guidance on dosing, potency, and thus expected efficacy… our study concluded that KinExA enables accurate measurement of the KD for strong protein–protein interactions (sub-nanomolar values) compared to FACS.”

— Vaish A. et al., 2020. Binding affinity determination of therapeutic antibodies to membrane targets: KinExA using cellular membranes for anti-CD20 antibody. Analytical Biochemistry, vol. 609

PROTEINS

“KinExA is particularly attractive to study protein–lipid interactions because native untagged molecules binding entirely in solution can be investigated.”

— Fleming J.K. et al., 2016. A novel approach for measuring sphingosine-1-phosphate and lysophosphatidic acid binding to carrier proteins using monoclonal antibodies and the kinetic exclusion assay. J. Lipid Research, 57(7), 1737–1747. doi:10.1194/jlr.D068866

SMALL MOLECULES

“The kinetics of binding and affinity of the 4 chimeric clones to human IL-21 were determined using a kinetic exclusion assay (KinExA)… Among the four lead clones, 19E3 had the highest affinity to recombinant human IL-21 of 243 fM.”

— Reena Varkey et al., 2019. Discovery and characterization of potent IL-21 neutralizing antibodies via a novel alternating antigen immunization and humanization strategy. PLoS ONE, 14(10): e0212236

DNA & APTAMERS

“Although numerous laboratory techniques have been developed to study the intricate interactions between proteins and DNA, each one of them has limited utility regarding applications. For better quantification of these important interactions, our work focused on using the Kinetics Exclusion Assay (KinExA) technology.”

— Leung E. et al., 2018. Quantifying protein–DNA interactions by kinetic exclusion assay. Biophysical Journal, 114(3): 442a
